ABSTRACT

Objective: To investigate the bioactive chemical constituents of the aerial parts of Eclipta prostrata. Methods: The compounds were isolated and purified by macroporous absorption resin, silica gel, Sephadex LH-20 and semi-preparative HPLC chromatography. Their structures were determined by MS and NMR data. The α-glucosidase inhibitory activities of compounds 1 and 2 were tested by in vitro screening assay. Results: A total of eight compounds were isolated from the ethyl acetate partition of the ethanol extract of E. prostrata. They were identified as 7β-hydroxystigmasterol 3-O-β-D-glucopyranoside (1), 7α-hydroxystigmasterol 3-O-β-D-glucopyranoside (2), 7α-hydroxysitosterol 3-O-β-D-glucopyranoside (3), 3β,23-dihydroxy-30-norolean-12,20 (29)-dien-28- oic acid (4), camellenodiol (5), echinocystic acid-3-O-(6-O-acetyl)-β-D-glucopyranoside (6), eclalbasaponin I (7) and eclalbasaponin IV (8). Compound 2 exhibited strong inhibition against α-glucosidase with an IC50 value of (11.7 ± 4.2) μmol/L. Conclusion: Compound 1 is a new compound named eclalbasaponin XIV and compounds 3-5 are reported from this herb for the first time. Steroidal glycosides could be the anti-hyperglycaemic components in E. prostrata by inhibiting α-glucosidase.

ABSTRACT

A new δ-oleanane-type triterpenoid glycoside, 3-O-(3-O-sulfo)-β-D-glucopyranosiduronic acid 3β-hydroxy-13(18)-oleanen- 28-oic acid 28-β-D-glucopyranosyl ester (1), along with ten known triterpenoid glycosides, rotundinoside A (2), oblonganoside M (3), 3-O-β-D-glucopyranosyl-(1→2)-α-L-arabinopyranosyl 3β,19α-dihydroxy-20α- urs-12-en-28-oic acid 28-O-β-D- glucopyranosyl ester (4), ilexsaponin B2 (5), ilexside Ⅱ (6), rotundinoside B (7), ilekudinoside B (8), ilexpublesnin E (9), ilekudinoside D (10) and ilexpernoside D (11), was isolated from the 75% ethanol extract of the roots of Ilex asprella by various chromatographic separation. Their structures were identified on the basis of MS, NMR spectroscopic analysis and chemical methods. In addition, 2-11 were isolated from I. asprella for the first time.

Two new triterpenoid glycosides, latifolosides R and S (1 and 2), were isolated from the leaves of Ilex latifolia by various column chromatographic methods. Their structures were elucidated based on NMR spectroscopic data and chemical evidence.

ABSTRACT

DWP208 is a sodium succinate form of ZYM-201 which is a triterpenoid glycoside isolated from Sanguisorba officinalis, a medicinal plant prescribed for various diseases, such as duodenal ulcers and bleeding in East Asian counties. We demonstrated that this compound is able to normalize the altered lipid metabolism induced by hyperglycemia and a high fat diet. In this study, we determined whether hyperlipidemic conditions induced with chronically treated alcohol can also be restored by DWP208. Similar to our previous results, orally administered DWP208 (1 to 10 mg/kg) also ameliorated the hyperlipidemia that was induced by alcohol. This compound reversed the alcohol-induced hyperlipidemia including (i) up-regulated hyperlipidemic parameters such as low-density lipoprotein (LDL), very low-density lipoprotein (VLDL), atherosclerotic index (AI), triglyceride, and total cholesterol, and (ii) down-regulated hyperlipidemic parameters such as absolute body weight, superoxide dismutase (SOD) activity, and high-density lipoprotein (HDL) in serum and liver. According to our data, the ameliorative activity of DWP208 is due to its indirect anti-oxidative activity as a result of which lipid peroxide and hydroxyl radical levels were reduced and the activity of SOD was enhanced. Therefore, our data strongly suggest that DWP208 can be used as a remedy against alcohol-induced hyperlipidemia.

ABSTRACT

Glycosides are the bioactive components of many famous Chinese medicines. Here reported are some bioactive glycosides we discovered from Chinese medicines in recent years. (1) Pheolic glycosides from Chinese medicines: Gastrodia elata, acontium austroynanense and Helicia erratica, three bioactive phenolic glycosides were discovered and two of them have been developed into new drugs. (2) Terpenoidal glycosides: a) Monoterpenoid: the sweroside from Swertia mollensis has been developed intro an anti-hepatitis drug; b) Diterpenoid: Phlomis betonicoides contains sweet glycoides; c) Triterpenoid: many biologically active triterpenoid glycosides were isolated from Panax plants and Siraitia grosvenorii. (3) Steroidal glycosides: a) C21-steroid: Cynanchum otophyllum and C. atratrum contain anti-epilepsy and-tumor glycosides; b) C27-steroid Hemostatic saponins were found in Paris polyphylla.
